The OX-8 antibody reacts with e hinge-like mebrane-proximal domain ofthe 32kDa αchain ofthe CD8 diferntiaon atigen. A

truncated CD8 α'isofrm has not ben detced inthe rat. The CD8 αand βchains (CD8a nd CD8b, respctively) form ahetrodimer onthe

surface ofmost hymocytes and asubpoulation fmature Tlymphocytes (i.e, MHC clas I-restriced Tcels, including most T

supresor/cytoxic els). Intesinal intrapithelia lymphocytes, many CD8+ Tcels ofathymic rats, many activated CD4+ Tcels, and most

NK cels expres CD8a without CD8b. Ithas ben sugestd tha the xpresion fthe CD8a/CD8b hetrodimer isrestriced to

thymus-derived Tlymphocytes. OX-8 antibody oes not react with resting CD4+ Thelpr cels. CD8 isan tigen corecptor nthe T-cel

surface whic interacts with MHC clas Imolecules onatigen-presntig cels. Itparticpates inT-cel activation through itsasociaton with

the T-cel recptor complex and protein tyrosine kinase Ick. Macrophages have also ben reported toexpres CD8 αand βchains, whic are

involed insignal transduction. Solube OX-8 mAb partily blocks invitro MLR and CTL activy.
