Background Information
SAP130, also known as Splicing factor 3B subunit 3, is part of the RNA splicing machinery. It is expressed by the Sf3b3 gene and encodes subunit 3 of the splicing factor 3b protein complex. As a component of this protein complex, it is associated with U2 snRNP and recruited to prespliceosomal complexes. Splicing factor 3b along with splicing factor 3a and a 12S RNA, form the U2 small nuclear ribonucleoprotein complex (U2 snRNP). In pre-mRNA’s, the splicing factor 3b/3a complex binds upstream of intron branch sites possibly anchoring the U2 snRNP to the pre-mRNA.
SAP130 is also a component of the STAGA (SPT3-TAF(II)31-GCN5L acetylase) transcription coactivator-HAT (histone acetyltransferase) complex, and the TFTC (TATA-binding-protein-free TAF(II)-containing complex). These complexes are believed to function in chromatin modification, transcription, splicing, and DNA repair.

FUNCTION: Subunit of the splicing factor SF3B required for 'A' complex assembly formed by the stable binding of U2 snRNP to the branchpoint sequence (BPS) in pre-mRNA. Sequence independent binding of SF3A/SF3B complex upstream of the branch site is essential, it may anchor U2 snRNP to the pre-mRNA. May also be involved in the assembly of the 'E' complex. Belongs also to the minor U12-dependent spliceosome, which is involved in the splicing of rare class of nuclear pre-mRNA intron by similarity.
