SRC-1 is a member of a gene family that functions in the full transcriptional activation of the steroid hormone receptor superfamily and contains intrinsic histone acetyltransferase activity. SRC-1 has a molecular weight of 160 kDa and has a basic helix-loop-helix (bHLH)-PAS domain.
Product Information
Format

Control
RIPA whole cell lysates from human A431, Jurkat cells, mouse 3T3/A31 cells or rat PC-12 cells Included Positive Antigen Control: Catalog # 12-301, Non-Stimulated A431 cell lysate. Add 2.5 μL of 2-mercaptoethanol/100 μL of lysate and boil for 5 minutes to reduce the preparation. Load 20 μg of reduced lysate per lane for minigels.
Presentation
Cultured supernantant in 0.05% sodium azide
Applications
Key Applications
Western Blotting
Immunoprecipitation
Application Notes
Western Blot Analysis: A 1:2000 to 1:8000 dilution of this lot detected Src in RIPA whole cell lysates from human A431 cells. Immunoprecipitation: 5-20 μL of a previous lot immunoprecipitated Src from 500 μg of A431 RIPA lysate.
Biological Information
Immunogen
KLH conjugated synthetic peptide (CKVADFGLARLIEDNEYTARQGA) of Src (avian sarcoma virus), residues 400-422. The immunogen sequence shares 22/23 amino acids with Yes and Fyn, and 21/23 amino acids with Lyn and Hck.
Clone
NL19
Host
Rabbit
Specificity
Src-tyrosine kinase family
Isotype
IgG
Species Reactivity
Rat Human Mouse
Species Reactivity Note
Predicted to cross react with frog and chicken. The immunogen sequence shares 22/23 amino acids with Yes and Fyn, and 21/23 amino acids with Lyn and Hck.
This gene is highly similar to the v-src gene of Rous sarcoma virus. This proto-oncogene may play a role in the regulation of embryonic development and cell growth. The protein encoded by this gene is a tyrosine-protein kinase whose activity can be inhibited by phosphorylation by c-SRC kinase. Mutations in this gene could be involved in the malignant progression of colon cancer. Two transcript variants encoding the same protein have been found for this gene.
SIZE:536 amino acids; 59835 DaSUBUNIT:Interacts with DDEF1/ASAP1 via its SH3 domain. Interacts with CCPG1 (By similarity). Interacts with CDCP1, PELP1, TGFB1I1 and TOM1L2.PTM:Phosphorylated on Tyr-530 by c-Src kinase (CSK). The phosphorylated form is termed pp60c-src. The phosphorylated tail interacts with the SH2 domain thereby repressing kinase activity.SIMILARITY:SwissProt: P12931 ## Belongs to the protein kinase superfamily. Tyr protein kinase family. SRC subfamily. & Contains 1 protein kinase domain. & Contains 1 SH2 domain. & Contains 1 SH3 domain.
