TNF receptor-associated factor 6 is a member of the TNF receptor associated factor (TRAF) protein family. TNF receptor-associated factor 6 proteins play an important role as adaptor protein for cell survival, proliferation, differentiation and stress responses. TNF receptor-associated factor 6 has unique binding behavior that result in signaling responses from the interleukin-1 receptor (IL-1R) toll-like receptor. TNF receptor-associated factor 6 mediates signaling indirectly with IL-1R/TLR through IRAK. This leads to activation of NF-κB and MAP kinase signaling pathways though downstream association with the TAB/TAK-1 complex. TNF receptor-associated factor 6 is also involved in the activation of Akt/PKB and serves as a direct E3 ligase which is essential for Akt ubiquitination.
