ErbB4/Her4 is a member of the ErbB receptor family of tyrosine kinases. It is largely expressed in many breast carcinoma cell lines; however, it is also found in normal skeletal muscle, heart, pituitary, brain, and cerebellum. This receptor is more specified than the others in this family because of its unique ability of the C-terminal to bind to a PDZ domain.

Presentation
Rabbit Monoclonal in buffer containing 50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl containing 40% Glycerol, 0.01% sodium azide and 0.05% BSA.
Applications
Application
This Anti-phospho-Her4/ErbB-4 (Tyr1258) Antibody, clone EP2272AY, Rabbit is validated for use in WB for the detection of phospho-Her4/ErbB-4 (Tyr1258).
Key Applications
Western Blotting
Biological Information
Immunogen
Phospho-specific peptide corresponding to residues surrounding Tyr1258 of human Her4/ErbB-4.
Epitope
Phosphorylated Tyr1258
Clone
EP2272AY
Host
Rabbit
Specificity
This antibody detects Her4/ErbB-4 phosphorylated on Tyr258.
FUNCTION:Specifically binds and is activated by neuregulins, NRG-2, NRG-3, heparin-binding EGF-like growth factor, betacellulin and NTAK. Interaction with these factors induces cell differentiation. Not activated by EGF, TGF-A, and amphiregulin. CATALYTIC ACTIVITY:ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate. SUBUNIT STRUCTURE:Homodimer or heterodimer with each of the other ERBB receptors Potential. Interacts with PDZ domains of DLG2, DLG3, DLG4 and the syntrophin SNTB2. Interacts with CBFA2T3, MUC1 and WWOX. SUBCELLULAR LOCATION:Membrane; Single-pass type I membrane protein. TISSUE SPECIFITIY:Expressed at highest levels in brain, heart, kidney, in addition to skeletal muscle, parathyroid, cerebellum, pituitary, spleen, testis and breast. Lower levels in thymus, lung, salivary gland, and pancreas. Isoform JM-A and isoform JM-B are expressed in cerebellum, but only the isoform JM-B is expressed in the heart. DOMAIN:The WW-binding motifs mediate interaction with WWOX. PTM:Isoform JM-A is processed but not isoform JM-B. So, they respectively represent cleavable and non-cleavable forms of the receptor.
Ligand-binding increases phosphorylation on tyrosine residues. SEQUENCE SIMILARITIES:Belongs to the protein kinase superfamily. Tyr protein kinase family. EGF receptor subfamily.
Contains 1 protein kinase domain.
Product Usage Statements
Quality Assurance
Evaluated by Western Blot on SKBR3 cell lysates.
Western Blot Analysis: A 1:1,000-5,000 dilution of this antibody was used to detect Her4/ErbB-4 on SKBR3 cell lysate.
Usage Statement
Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage Conditions
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
