SOCS1, also known as Suppressor of cytokine signaling 1, or JAK-binding protein (JAB), STAT-induced STAT inhibitor 1 (SSI-1), or Tec-interacting protein 3 (TIP3), and encoded by the gene SOCS1/CISH1/SSI1/TIP3, is involved in the negative regulation of cytokines signaling that is mediated by several kinases including JAKs, TEC, KIT, GRB2 and VAV kinases. SOCS1 inhibits kinase activity via binding to the target kinase. SOCS1 also appears to be a major regulator of signaling by interleukin 6 (IL6) and leukemia inhibitory factor (LIF), and SOCS1regulates interferon-gamma mediated sensory neuron survival and is a negative regulator in IGF1R signaling pathway. Finally, SOCS1 forms the substrate recognition component of an ECS (Elongin BC-CUL2/5-SOCS-box protein) E3 ubiquitin ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. SOCS1 is localized to the nucleus and cytoplasmic vesicles. SOCS1 is expressed in all tissues with high expression in spleen, small intestine and peripheral blood leukocytes.
