HCA149A recognizes human DJ-1, also known widely as PARK7, a nuclear and cytoplasmic protein of approximately 21kDa belonging to the C56 peptidase family.
DJ-1 plays roles in transcriptional regulation and reaction to anti-oxidative stress. Research indicates that DJ-1 is responsible for protecting neurones against oxidative stress and cell death. De-regulation of DJ-1 oxidation appears to be connected to the onset of conditions such as Parkinson's Disease.
Research also suggests that the oxidation status of DJ-1, particularly at C106, modulates the function of DJ-1, AbD Serotec have also introduced an antibody that specifically recognises DJ-1 when oxidized at C106 (product code HCA124).
Product Form
A bivalent human recombinant Fab selected from the HuCAL® phage display library, expressed inE. coli. This Fab fragment is bivalent by dimerization of the bacterial alkaline phosphatase fusion protein. The antibody is tagged with V5 and a Strep-tag at the C-terminus of the antibody heavy chain. This antibody is supplied lyophilised.
Storage
Prior to reconstitution store at +4oC.
After reconstitution store at -20oC.
Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life
12 months from date of reconstitution.

Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.
This product does not require protein digestion pre-treatment of paraffin sections. This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.
Sodium citrate buffer pH 6.0 is recommended for this purpose.
