HCA063 recognises human cyclin-dependent kinase 2 (Cdk2), a member of the serine/threonine kinase family of cyclin-dependent kinases. Cyclin-dependant kinases (CDKs) are a class of enzymes that control cell cycle progression in a variety of organisms. Cdk2 forms active heterodimeric complexes with regulatory subunits, including cyclin A or cyclin E, resulting in the phosphorylation of the retinoblastoma tumour suppressor protein. Its activity is regulated by phosphorylation and CDK inhibitors p21/WAF1 (CDKN1A) and p27/kip1 (CDKN1B).
Cdk2 is involved in the regulation of G1/S transition and S phase progression of the cell cycle but may also play a role in apoptosis and DNA double strand break repair following ionising radiation.
Cdk2 can be up-regulated in tumours, including adenomas and breast cancers, particularly in conjunction with cyclin over-expression.
