对于Antibody Pair的特异性和灵敏度，请参照相关的PathScan® Sandwich ELISA Kit。注意：经内部测试确认，本抗体对可以检测到所列物种中的蛋白。但对其他物种中的同源蛋白可能也可以检测。CST的PathScan® Total Src Sandwich ELISA Antibody Pair是可以替代PathScan® Total Src Sandwich ELISA Kit #7984的更经济的产品。其中提供了捕获抗体、检测抗体(100X stocks)和HRP偶联的二抗(1000X stock)。这些试剂足够完成4 x 96 孔板的ELISAs实验。在96孔板中，加入Src 捕获抗体进行包被。封闭后，加入细胞裂解液，随后分别加入Src捕获抗体、anti-mouse IgG的HRP偶联抗体。最后加入HRP底物—— TMB，进行显色。产生的吸光值与Src蛋白的数量成正比。Src家族蛋白酪氨酸激酶包括Src, Lyn, Fyn, Yes, Lck, Blk, Hck, 对调节真核细胞的生长和分化很重要(1)。Src的活性是通过两个酪氨酸位点的磷酸化来调控的，但是这两者的调控效应相反。激酶结构域的活化环中的Tyr416位点的磷酸化，能够上调酶活性；而蛋白羧基端尾部Tyr527位点被Csk磷酸化后，则会使酶的活性降低(2)。

Description

Cell Signaling Technology's PathScan® Total Src Sandwich ELISA Antibody Pair is being offered as an economical alternative to our PathScan® Total Src Sandwich ELISA Kit #7984. Capture and detection antibodies (100X stocks) and HRP-conjugated secondary antibody (1000X stock) are supplied. Sufficient reagents are supplied for 4 x 96 well ELISAs. The Src rabbit capture antibody is coated in PBS overnight onto a 96 well microplate. After blocking, cell lysates are added followed by a Src mouse detection antibody and an anti-mouse IgG, HRP-linked antibody. HRP substrate (TMB) is then added for color development. The magnitude of the absorbance for this developed color is proportional to the quantity of Src protein.

Background

The Src family of protein tyrosine kinases, which includes Src, Lyn, Fyn, Yes, Lck, Blk, and Hck, are important in the regulation of growth and differentiation of eukaryotic cells (1). Src activity is regulated by tyrosine phosphorylation at two sites, but with opposing effects. While phosphorylation at Tyr416 in the activation loop of the kinase domain upregulates enzyme activity, phosphorylation at Tyr527 in the carboxy-terminal tail by Csk renders the enzyme less active (2).
