PLD2.Phospho-PLD1 (Thr147)抗体识别内源性的Thr147磷酸化的PLD1蛋白。此抗体不与磷酸化的PLD2蛋白交叉反应。该多克隆抗体用与人类PLD1中Thr147位点附近的氨基酸序列对应的人工合成磷酸化的肽段免疫动物而制成。该抗体使用蛋白A和蛋白亲和层析纯化而得。卵磷脂特异型磷酸酶D（PLD），水解卵磷脂（PC）产生胆碱和磷脂酸（PA）。PA是第二信使甘油二酯（DAG）的前体。PLD的两种亚型（PLD1和PLD2）目前已被确认。两者都被蛋白激酶、小GTP酶和Ca2+调控（1）。PLD1可以被PKC在Ser2、Ser561和Thr147位点磷酸化（2,3）。磷酸化Thr147和Ser561调节PLD1的活性（3）。

Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic phosphopeptide corresponding to residues surrounding Thr147 of human PLD1. Antibodies are purified by protein A and peptide affinity chromatography.

Background

Phosphatidylcholine-specific phospholipase D (PLD) hydrolyzes phosphatidylcholine (PC) to produce choline and phosphatidic acid (PA). PA is the precursor of the second messenger, diacylglycerol (DAG). Two isoforms of PLD (PLD1 and PLD2) have been identified so far. Both are regulated by protein kinases, small GTPases and Ca2+ (1). PLD1 is phosphorylated at Ser2, Ser561 and Thr147 by PKC (2,3). Phosphorylation at Thr147 and Ser561 regulates PLD1 activity (3).
