Survivin (71G4E) Rabbit mAb (Alexa Fluor®647 Conjugate) detects endogenous levels of total survivin protein.
Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding cysteine 60 of human survivin. The antibody was conjugated to Alexa Fluor®647 under optimal conditions with an F/P ratio of 2-6. The Alexa Fluor®647 dye is maximally excited by red light (e.g. 633 nm He-Ne laser). Antibody conjugates of the Alexa Fluor®647 dye produce bright far-red-fluorescence emission, with a peak at 665 nm.
Description
Cell Signaling Technology antibody is conjugated to Alexa Fluor®647 fluorescent dye and tested in-house for direct flow cytometric analysis of human cells. The unconjugated antibody, #2808 reacts with survivin from human, mouse, and rat. CST expects that Survivin (71G4E) Rabbit mAb (Alexa Fluor®647 Conjugate) will also recognize survivin in these species.
Background
Survivin is a 16 kDa anti-apoptotic protein highly expressed during fetal development and cancer cell malignancy (1). Survivin binds and inhibits caspase-3, controlling the checkpoint in the G2/M-phase of the cell cycle by inhibiting apoptosis and promoting cell division (2,3). This regulatory process requires the phosphorylation of survivin at Thr34 by p34 cdc2 kinase (4). Gene targeting using a Thr34 phosphorylation-defective survivin mutant, as well as antisense survivin, have been shown to inhibit tumor growth (5,6).
Storage: Supplied in PBS (pH 7.2), less than 0.1% sodium
azide and 2 mg/ml BSA. Store at 4°C. Do not aliquot the
