Mouse IL-17 mRNA is specifically expressed by activated alpha beta TCR + CD2-CD8-T-cells, a small subset with a potentially important role in immune regulation. IL-17 induces the secretion of IL-6, IL-8, PGE2, MCP-1 and G-CSF by adherent cells like fibroblasts, keratinocytes, epithelial and endothelial cells. IL-17 is also able to induce ICAM-1 surface expression of T-cells. IL-17 has been shown to support the growth of hemopoietic progenitors, it has stimulatory effects on granulopoiesis in mice.
IL-17 may affect osteoclastic resorption and thereby mediate bone destruction accompanying some inflammatory diseases. Astrocytes produce NO when stimulated by IL-17, which suggests a possible role for IL-17 in the inflammatory diseases of the CNS. Forced expression of murine IL-17 induces immune responses and multiorgan inflammation.
Interleukin-17 is involved in the regulation of hematopoiesis and its effects are dependent on the physiological/pathological status of the organism. IL-17 expression in airways is upregulated upon allergen exposure like in allergic asthma. Its crucial role in T-cell activation suggests that IL-17 is involved in the development of autoimmune arthritis.
