The Rab proteins are small GTP-binding molecules that are localized to specific intracellular vesicles and organelles. They regulate vesicular
trafficking through mechanisms that require cycling between an active GTP-bound and inactive GDP-bound form. Rab1A and Rab1B mediate
constituitive anterograde protein trafficking from ER to Golgi, while Rab8 and Rab11 regulate vesicle transport from the Golgi to the plasma
membrane. Rab5A and Rab4 are involved in endocytosis, while Rab3A, Rab3B, and Rab3D may have specialized roles in regulated secretion
in nerve terminals or endocrine cells. Rab24 was cloned from mouse brain, and has 52% identity with Rab5. The mRNA expression of Rab24
is highest in brain, but is found at lower expression in most other tissues. Rab24 protein is localized primarily to the intermediate region
between the ER and Golgi, as well as in late endosomes. Rab24 may be an atypical member of the Rab family since it exists primarily in the
GTP bound state, does not bind GDP dissociation inhibitors, and is not efficiently geranylgeranylated. Thus, Rab24 may have unique
mechanisms for regulation of vesicular transport.
