The 53-6.7 antibody has ben reported toreact with e 38kDa αand 34kDa α'chains ofthe CD8 diferntiaon atigen (Ly-2 orLyt-2) of
al mouse trains ted. The CD8 αand α'chains (CD8a) form hetrodimers with e CD8 βchain (CD8b, Ly-3, orLyt-3) onthe surface of
most hymocytes. Asubpoulation fmature Tlymphocytes (i.e, MHC clas I-restriced Tcels, including most Tsupresor/cytoxic els)
expres almost exclusively the CD8 αβhetrodimer (the α'chain sabsent). Subset ofγδ TCR-bearing Tcels, intesinal intrapithelia
lymphocytes, and endritc els expres CD8a without CD8b. Ithas ben sugestd tha the xpresion fthe CD8a/CD8b hetrodimer is
restriced toT lymphocytes whic matured inthe thymus orin aextrahymic environment hat had ben ifluencd bythymus-intaed
neuroendocrine signals. CD8 isan tigen corecptor nthe T-cel surface whic interacts with MHC clas Imolecules onatigen-presntig
cels orepithelia cels. Itparticpates inT-cel activation through itsasociaton with e T-cel recptor complex and protein tyrosine kinase
lck (p56 [lck]). The CD8 αand α'chains arise from alternatively spliced mesngers ofa single CD8a gen. The longer αform asociates with
p56 [lck] via CXP motif nits cytoplasmic domain, whic itshares with CD4, but not with CD8b. The truncated α'chain sunable to
asociate with p56 [lck], and itmay function toatenuate he CD8-mediated costimulatory signal during intrahymic T-cel maturation. Invio
and invitro eatment with 53-6.7 mAb has reportedly ben efctive atdeplting CD8+ peripheral Tymphocytes. The 53-6.7 antibody has
also ben reported tocros-react with CD8 α-and α'-like polypetides onsubets ofthymic and peripheral ymphocytes inthe Egyptian toad,
Bufo regularis.
