Goat anti Human SMAD2 antibodyrecognizes human SMAD2, also known as MADH2 or Mothers against decapentaplegic homolog 2. SMAD2 is a 467 amino acid ~50kDa intracellular signaling protein responsible for mediating the signal from TGFβ. SMAD2 associates woth the TGFβ receptor via the SMAD anchor for receptor activation (SARA) and on binding of TGFβ to the receptor SMAD2 is phosphorylated and dissociates from SARA. SMAD2 then forms an association with another SMAD family member SMAD4, important for translocation to the nucleus where it binds to its target promoters and activates TGFβ induced pathways.
There are two identified isoforms of SMAD2 with the short isoform lacking an internal sequence from amino acid 79 to amino acid 108. The immunogen sequence for Goat anti Human SMAD2 antibody is contained within this region and hence this antibody is only expected to recognize the long, canonical isoform of SMAD2..
SMAD2 is expressed at high levels in placenta skeletal muscle and heart (Xuanet al.2007)

Antiserum Preparation
Antisera to MADH-2 were raised by repeated immunisations of goats with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.
