AHP2253 specifically recognises an epitope within the N-Terminal (NT) region of human MAML1 (Mastermind-like Protein 1), a widely expressed nuclear protein and member of the mastermind family, which acts as a crucial coactivator of Notch-mediated transcriptional activation, and plays a role in hematopoietic development, by acting as a regulator of Notch-mediated lymphoid cell fate decisions.
MAML1 interacts with the Mediator complex component CDK8 (Cyclin-dependent kinase 8), enhancing the phosphorylation and proteolytic turnover of the Notch intracellular domain (NICD) within the nucleus, and forms a DNA-binding complex with RBPSUH/RBP-J kappa/CBF1, playing a vital role in the transcriptional activation of Notch targets. Studies have shown that the NT region of MAML1 interacts with the DNA-binding domain of the tumor suppressor protein p53, and that over-expression of MAML1, greatly increases p53-mediated gene transcription and biological function.
