AHP1573 specifically recognises human LEF1 (lymphoid enhancing-binding factor 1), a nuclear protein and member of the TCF/LEF transcription factor family, developmentally regulated and expressed in pre-B and T lymphocytes, which binds to a functionally active site on the T-cell antigen receptor (TCR) alpha enhancer and plays a vital role in the Wnt/Wingless signalling pathway.
The interaction of full-length LEF1 with beta-catenin acts as a Wnt signal transducer, which can be blocked by truncated LEF1 isoforms which do not bind beta-catenin and occupy Wnt target genes. Cancer studies have indicated that during tumour progression, the relative levels of the LEF isoforms may vary compared to that of normal tissue, thereby influencing the effectiveness of Wnt signalling within the nucleus.
