AHP1425 recognises Human Rad52, a protein which functions in DNA repair through the homologous recombination of DNA double strand breaks (DSBs). DSBs can be generated through exposure to ionizing radiation or by endogenous cellular reactions, and Rad52 acts during the latter stages of recombination after the DSBs are processed into single stranded DNA (ssDNA).
Rad52 functions together with Rad51 and RPA (Replication Protein A), where it has two distinct activities. First, Rad52 and RPA form a co-complex on ssDNA, where Rad52 functions to recruit Rad51. In addition, Rad52 has ssDNA binding and annealing activities, particularly ssDNA saturated with RPA.
Rad52 is present at low levels during the G1 phase of the cell cycle, when recombinatorial repair is absent. The level rises steadily through S phase reaching a maximum in G2 phase when homologous repair is also at its highest level.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.Sodium citrate buffer pH 6.0 is recommended for this purpose.

Antiserum to human RAD52 was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.
