AHP1227 detects an epitope within the N-terminal region of human CD61, also known as integrin beta 3 (ITGB3). Integrins are integral cell-surface proteins composed of an alpha chain and a beta chain. CD61 is a 90-110 kDa glycoprotein which binds to CD41 (ITGA2B) to form the GPIIb/IIIa complex, and CD51 (ITGAV) to form the vitronectin receptor.
Binding of fibrinogen to the GPIIb/IIIa complex brings about platelet/platelet interaction, leading to rapid platelet aggregation and clotting. The vitronectin receptor is essential for endothelial cell migration, proliferation, and prevention of apoptosis.
Elevated expression of the vitronectin receptor is associated with tumour progression and metastases formation on melanoma cells, and is used by human cytomegalovirus (HCMV) as a co-receptor to initiate viral entry and signalling.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.Sodium citrate buffer pH 6.0 is recommended for this purpose.
