AHP1190 specifically recognises an epitope within the C-Terminal (CT) region of nicastrin, a widely expressed type I transmembrane glycoprotein and member of the nicastrin family, which is constitutively expressed by neuronal cells, and is an essential component of the endoprotease gamma-secretase complex.
The gamma-secretase complex, consisting of APH-1 (APH-1A or APH-1B), presenilin-1 or -2 (PSEN1 or PSEN2), nicastrin (NCSTN), and Pen-2 (PEN2), plays a vital role in the evolutionarily conserved Notch signaling pathway, by catalyzing the intramembrane S3 cleavage of ligand-activated Notch receptor proteins (Notch1-4), resulting in the generation and release of an activated NICD fragment. Nicastrin is also involved in the cleavage of the beta-amyloid precursor protein, to form the amyloid beta peptide, a major component of the neuritic plaques found in Alzheimer’s disease.
