AHP1023 recognises human TOLLIP or Toll interacting protein. TOLLIP is a 30kDa adaptor molecule for Toll-like receptors, transducing signals and activating kinases and transcription factors in response to an infection. It is thought to inhibit IRAK-1 activity. This facilitates the termination of the IL-1RI-, TLR2- and TLR4-triggered, NF-kB and JNK signalling pathways during inflammation. Furthermore, TOLLIP is also thought to form a complex with Tom1, which regulates endosomal trafficking of ubiquitinated proteins. Unlike most other inflammation suppressors, TOLLIP is constitutively expressed.
