PGP 9.5 hydrolyses a peptide bond at the C-terminal glycine of the proteolysis tagging protein ubiquitin, thereby generating/recycling ubiquitin monomers, essential in the ubiquitin-proteasome cytoplasmic protein degradation system. PGP 9.5 is a focus area for neurological disorders such as Alzheimer’s and Parkinson’s disease and has been linked with tumor invasion and progression.
